Chemoprotective effect of cysteamine against the induction of micronuclei by methyl methanesulfonate and cyclophosphamide Genet. Mol. Biol.
Santos-Mello,Renato; Deimling,Luiz Irineu; Lauer Júnior,Claudio; Carvalho,Thaís Rieger de.
Cysteamine or 2-mercaptoethylamine (MEA) is an aminothiol with a well-known radioprotective action. No specific information is available in the literature about the possible chemoprotective action of MEA against genotoxic chemical agents. This paper presents the results of studies on the ability of MEA to protect mouse bone marrow polychromatic erythrocytes against the induction of micronuclei by alkylating agents such as methyl methanesulfonate (MMS) and cyclophosphamide (CP). We observed that MEA administered intraperitoneally 30 min before or 30 min after the administration of MMS or CP significantly reduced the frequency of micronucleated polychromatic erythrocytes (MNPCEs) induced by the alkylating agents. When MEA was administered in combination with...

Colocalization of coilin and nucleolar proteins in Cajal body-like structures of micronucleated PtK2 cells BJMBR
Silva,N.P.; Christofolini,D.M.; Mortara,R.A.; Andrade,L.E.C..
Cajal bodies (CB) are ubiquitous nuclear structures involved in the biogenesis of small nuclear ribonucleoproteins and show narrow association with the nucleolus. To identify possible relationships between CB and the nucleolus, the localization of coilin, a marker of CB, and of a set of nucleolar proteins was investigated in cultured PtK2 cells undergoing micronucleation. Nocodazol-induced micronucleated cells were examined by double indirect immunofluorescence with antibodies against coilin, fibrillarin, NOR-90/hUBF, RNA polymerase I, PM/Scl, and To/Th. Cells were imaged on a BioRad 1024-UV confocal system attached to a Zeiss Axiovert 100 microscope. Since PtK2 cells possess only one nucleolus organizer region, micronucleated cells presented only one or...

Kirschbaum,Aline A.; Seriani,Robson; Pereira,Camilo D. S.; Assunção,Andrea; Abessa,Denis M. de Souza; Rotundo,Matheus M.; Ranzani-Paiva,Maria J. T..
The aquatic environment receives many contaminants that can induce damages at the molecular, biochemical, cellular and physiological levels. Centropomus parallelus , an important food resource for local populations, is a predator fish that feeds on small fishes and benthic invertebrates, thus being vulnerable to the bioconcentration and biomagnification processes. This study aimed to evaluate cytogenotoxic responses in erythrocytes from C. parallelus juveniles collected in the Cananéia and São Vicente estuaries, both in winter and in summer. After anesthesia, blood samples were collected by caudal puncture. Blood smears were prepared on glass slides and stained with May-Grünwald-Giemsa dye. Two thousand cells were analyzed per slide (1000x), and nuclear...

Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG
Andrioli,Nancy B; Mudry,Marta D.
The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml. An increase was also observed in the rate of metaphase and anaphase, which was, in turn, indicative of alterations in chromatid...

Effects of cadmium on root growth, cell division and micronuclei formation in root tip cells of Allium cepa var. agrogarum L Phyton
Wang,QL; Zhang,LT; Zou,JH; Liu,DH; Yue,JY.
The effects of various cadmium (Cd) concentrations (10, 50 and 100 μM) on root growth, cell division and micronuclei formation in root tip cells of Allium cepa var. agrogarum L. were investigated to better understand the processes of Cd-induced apoptosis. The results indicated that 10 μM of Cd had an obviously passive influence on root growth during the 24 h treatment, and that the influence was even more serious with increasing Cd concentrations and duration of treatments. The mitotic index decreased with increasing Cd concentration and duration of treatments. Cadmium induced c-mitosis, chromosome bridges, chromosome stickiness and micronuclei. The frequency of cells with these disorders varied with prolonging the duration of treatments...

Genetic stability in synthetic wheat accessions: cytogenetic evaluation as a support in breeding programs Ciência Rural
Frizon,Patrícia; Brammer,Sandra Patussi; Lima,Maria Imaculada Pontes Moreira; Castro,Ricardo Lima de; Deuner,Carolina Cardoso.
ABSTRACT: Synthetic wheat is developed by crossing tetraploid species ( Triticum turgidum , AABB) with a diploid species ( Aegilops tauschii , DD), followed by chromosome duplication through the use of colchicine to restore the resultant sterile hybrid to a fertile hexaploid plant. The main importance of producing synthetically improved wheat is to increase their genetic variability and to incorporate genes that code for resistance to biotic and abiotic stressors. This study aimed to evaluate the presence of micronuclei (MN) and the meiotic index (MI) in the tetrad phase in synthetic wheat accessions and cultivars ( Triticum aestivum ) stored at the Germplasm Bank of Embrapa Trigo (Brazil), in order to identify and select genetically stable accessions for...

Poli,M.N; López Miranda,L.A; Fernández Iriarte,P.J; Zanier,G.J; Iúdica,C.E.
Previous research has shown connections between the exposure to different agents and mutations in germinal and somatic cells. One of such agents, nitrate (NO3), can be potentially genotoxic for individuals who drink high nitrate well water, and may cause a subsequent impact on their health and on the whole ecosystem. The aim of this research was to conduct genotoxicity assays in somatic cells from people living in the northern area of Mar del Plata city, Argentina. The purpose of these experiments was, in turn, to establish the possible relationships between mutagenic effects and high nitrate water consumption. To this end, different diagnostic tests were carried out to detect potential genetic and/or chromosomal alterations. A non-exposed population equal...

Santos,Celina Alcoforado; Novaes,Larissa Simões; Gomes,Levy Carvalho.
The high toxicity of diesel components makes oil spills a threat to the biota in coastal marine environments. The genotoxic effect of the diesel water-soluble fraction (DWSF) on Hippocampus reidi (Ginsburg, 1933) was assessed. Fish were exposed to three different DWSF dilutions for up to 96 hours, and genotoxicity was analyzed using the micronuclei test and the comet assay. The micronuclei test revealed no significant differences between any of the DWSF dilutions and the control in the 24-hours period; however, micronuclei increased in fish exposed to 1:500 and 1:100 DWSF dilutions for 96 hours. For all dilutions, there was an increase in micronuclei in fish exposed for 96 hours when compared to those exposed for 24 hours. The tested dilutions increased...

Genotoxicity of titanium dioxide nanoparticles and triggering of defense mechanisms in Allium cepa Genet. Mol. Biol.
Santos Filho,Ronaldo dos; Vicari,Taynah; Santos,Samuel A.; Felisbino,Karoline; Mattoso,Ney; Sant’Anna-Santos,Bruno Francisco; Cestari,Marta Margarete; Leme,Daniela Morais.
Abstract Titanium dioxide nanoparticles (TiO2NPs) are widely used and may impact the environment. Thus, this study used a high concentration of TiO2NP (1000 mg/L) to verify the defense mechanisms triggered by a plant system – an indicator of toxicity. Furthermore, this study aimed at completely characterizing TiO2NP suspensions to elucidate their toxic behavior. TiO2NPs were taken up by meristematic cells of Allium cepa, leading to slight inhibition of seed germination and root growth. However, severe cellular and DNA damages were observed in a concentration-dependent manner (10, 100, and 1000 mg/L). For this reason, we used the highest tested concentration (1000 mg/L) to verify if the plant cells developed defense mechanisms against the TiO2NPs and...

Induced CYP1A activity and DNA damage in fish from the middle Paraíba do Sul River basin, southeastern Brazil Biological Sciences
Morado, Claudio Nona; Martins, Thiago Estevan Parente; Araujo, Francisco Gerson; Paumgarten, Francisco José Roma; Gomes, Iracema David.
The Paraíba do Sul River (PSR) drainage basin in Southeastern Brazil covers one of the most industrialized and densely populated regions of the country. The impact of chemical contamination on the PSR basin seems to be more pronounced in its middle reach where a number of potentially polluting plants are located. In this study, we used hepatic EROD activity - a biomarker of exposure to CYP1A-inducing pollutants (e.g. PAHs, PCDD/Fs, PCBs) - and the incidence of micronucleated erythrocytes (Mn) in the peripheral blood - a biomarker of effects of DNA-damaging agents (e.g. PAHs) - to evaluate the effects of pollution on two native fish species, Geophagus brasiliensis and Pimelodus maculatus. Results showed that the incidence of Mn and EROD in G. brasiliensis...
